Galactomannans of Gleditsia triacanthos, Caesalpinia pulcherrima and Adenanthera pavonina were structurally and thermally characterized. Methylation analyses confirmed that these three galactomannans are composed by a 4-linked mannose polymer with galactose side chains attached at the C6 position. G. triacanthos galactomannan had an estimated average degree of polymerization of 224 mannose residues, with a degree of branching of 0.24, C. pulcherrima galactomannan had a degree of polymerization of 252 and a degree of branching of 0.30, and A. pavonina galactomannan had a degree of polymerization of 475 and a degree of branching of 0.60. Enzymatic hydrolysis with endo-ß-mannanase and the subsequent analyses of the low molecular weight fraction by ESI-MS/MS indicated the presence of characteristic structural features of galactomannans. Furthermore, acetyl and pentosyl residues were detected in the galactomannan of G. triacanthos and analyses of higher molecular weight fractions from G. triacanthos were performed, confirming that presence. Differential scanning calorimetry (DSC) showed the presence of two peaks related with water loss, confirmed by thermogravimetric analysis (TGA). The different compositions of galactomannans with different mannose content and different molecular weights influence their thermal behaviour. A higher content of mannose lead to higher values of the enthalpy change (first thermal transition) and glass transition temperature (Tg), while the enthalpy change of the second thermal transition was influenced by the viscosity average molecular weight (Mv) of the samples. This work provides understanding of structural and thermal properties of three galactomannans from non-conventional sources, thus contributing to a better insight on their possible food, pharmaceutical or biomedical applications.

Galactomannans from locust bean gum (LBG) are polysaccharides containing single Galp residues as side chains in a ß-(1¿4)-Manp backbone. Nevertheless, sugars other than mannose and galactose are always detected, namely, arabinose and glucose, although in small amounts. In order to demonstrate that LBG galactomannans contain other structural features than those usually reported in literature, the galactomannans from LBG obtained from two different commercial sources were submitted to a selective degradation by Aspergillus niger endo-ß-d-mannanase and the oligosaccharides (OS) obtained were fractioned by size exclusion chromatography using a Biogel-P2 (100-1800 Da). A. niger endo-ß-d-mannanase is able to hydrolyse the ß-(1¿4)-linked mannan backbone when the mannose residues are not hindered by the presence of substituents. This enables collection of fragments of the substituted mannan polysaccharide in the different chromatographic fractions that can be analysed by Electrospray Tandem Mass Spectrometry (ESI-MSn). Both samples showed the presence of hexose OS (Hex3-6), acetylated hexoses (AcHex3-6), hexoses substituted with a pentose unit (PentHex3-6), and OS composed by acetylated hexoses substituted with a pentose unit (PentAcHex3-6). These structures were confirmed by GC-qMS of reduced and permethylated oligosaccharides. Sugar and glycosidic-linkage analysis allowed to assign the pentose unit to a terminally-linked arabinofuranose residue at the O-6 of a (1¿4)-Manp residue. The presence of arabinose substituted OS and acetylated mannoses have never been reported to occur in LBG. .

The galactomannans purified from coffee infusions have been shown to present in vitro immunostimulatory activity on murine B- and T-lymphocytes. These properties have also been shown characteristic of the galactomannans recovered from coffee residue by strong alkali solutions and rendered soluble in water by partial acetylation. In this study, purified fractions of galactomannans with immunostimulatory activity obtained from coffee infusion and from coffee residue were compared according to their average molecular weight determined by size-exclusion chromatography on Sephacryl S300, glycosidic-linkage composition by methylation analysis, selective hydrolysis by an endo-(1 ¿ 4)-ß-d-mannanase, analysis of the resultant oligosaccharide profile by Bio-Gel P2 separation, and determination of the pattern of acetylation by electrospray tandem mass spectrometry (ESI-MS/MS). It was found that the galactomannans have a comparable molecular weight (90–110 kDa), and similar glycosidic-linkage composition. However, the galactomannans from coffee residue were preferentially acetylated in the side chain residues whereas the galactomannans recovered from coffee infusions only had acetyl groups directly linked to the backbone residues. These results show that these polysaccharides that present comparable immunostimulatory properties have different acetylation patterns.

Coffee infusion mannans are acetylated polysaccharides containing single Galp and Araf residues as side chains of a beta-(1 --> 4)-Manp backbone. These mannans are structurally similar to the bioactive acetylated mannans from Aloe vera (AV). In this study, acetylated mannans were obtained from two coffee infusions prepared from light and dark roasted beans. These samples were tested for their immunostimulatory activity and compared with an extract of AV mannan and with locust bean gum (LBG) galactomannans. The coffee samples, as well as the AV extract, stimulated murine B- and T-lymphocytes, as evaluated by the in vitro expression of the surface lymphocyte activation marker CD69, more marked on B- than on T-lymphocytes. In coffee samples, contrarily to the AV, no proliferative effect was noticed. LBG sample did not show any immunostimulatory activity. Because the material that remains in the residue of the hot water extraction was still very rich in mannans, a sequential extraction was performed and a main fraction was recovered with a 4 M NaOH solution. Because this material was insoluble in water, a partial acetylation was performed. These polysaccharides also showed immunostimulatory activity, opening the possibility of exploitation of coffee infusion and coffee residue as sources of bioactive polysaccharides.

The possibility of discrimination of the anomeric configuration (a or ß) of underivatized reducing glucopyranosyl-glucose disaccharides, using a hybrid mass spectrometer Q-TOF 2 (Micromass), a linear ion trap LXQ (Thermo), and a triple quadrupole Quattro (Micromass) with an electrospray source (ESI) was investigated. Differences observed in the relative abundances of specific product ions obtained from collisionally induced dissociation of the [M + Li]+ adducts were statistically analyzed, and discriminant analysis was performed. MANOVA has shown that anomeric configuration has influence on the combined dependent variables (relative abundances of m/z product ions) in all the three mass spectrometers used (Q-TOF 2, LIT, and QqQ). Discriminant analysis has shown that, in all instruments, it is possible to discriminate anomeric configurations and to build a diagnostic model. These diagnostic differences are even more relevant considering that no derivatization procedures are needed for obtaining this structural information. The Q-TOF 2 instrument has been shown to give data that allowed us to build a model with better discriminant power (Wilks' ¿ value of 0.014) followed by the QqQ instrument (Wilks' ¿ value of 0.029) and the LIT instrument (Wilks' ¿ value of 0.037).
