with ammonium. This is a costly process, consuming twelve to seventeen grams of carbohydrate per gram of N2 fixed. Although
its cost, it’s an important process, primarily for amino acid production and thereby the high nutritious value of these plants.
In this study we use Lotus japonicus as a model plant due to its capacity to form determinate nodules when inoculated with
Mesorhizobium loti (MAFF 303099) and genomic sequence information is available for both of the symbiotic partners. We intend
to analyze nodules collected five weeks after inoculation with bacteria. We will concentrate on the cytosolic fraction, where
the enzymes responsible for the supply of carbohydrates and the assimilation of nitrogen are expected to be located. The methods
used to study the proteins will be 2D gel electrophoresis combined with tandem mass spectrometry. While awaiting the formation
of the above mentioned nodules, a preliminary 2D gel analysis
were made with extracts of root nodules from a commercial variety of Lotus corniculatus cv. Trueno. In the first analysis
nine spots were chosen at random and eight annotated proteins were identified: Three where enzymes from the glycolytic pathway:
fructokinase like-protein, phosphoglyceromutase and enolase, two isoenzymes of glutamine synthetase used in nitrogen assimilation,
together with an actin and a peroxidase, all of them of plant origin. An S-adenosylmethionine synthase was also identified,
but of Rhizobial origin, indicating
breakage of bacteroids in the isolation procedure. One spot could not be identified. Upon analysis and identification of many
more spots we hope to be able to compare data from Lotus nodules with published data from soybean (determinate nodules) and
Medicago truncatula (indeterminate nodules) in order to reveal major metabolic differences between the two types of nodules. |
