Linhas de Investigação (Research fields)
1. Role and regulation of Glutamine synthetase (GS) in Medicago truncatula
Objectivos»Goals: (i) Characterize the entire GS gene family in Medicago truncatula (ii) Analyze patterns of gene and protein expression (iii) Determine the catalytic properties of each isoenzyme.
2. Transcript expression profilling of root nodules with altered levels of GS activity
Objectivos»Goals: (i) Transcriptome analysis of root nodules with inactive GS (ii) Identification of genes associated with GS activity .
3. Post-translational regulation of Glutamine Synthetase (GS) by ubiquitin and/or ubiquitin-like proteins
Objectivos»Goals: (i) Characterize the putative post-translational regulation of GS by ubiquitin and/or ubiquitin like proteins (ii) investigate the functional relevance of these modifications for nitrogen assimilation in Medicago truncatula.
4. Nitrogen signaling and plant development in Medicago truncatula
Objectivos»Goals: It is well known that microbes are able to modulate plant development. An important example is the interaction of legumes with nitrogen-fixing bacteria (Rhizobium), which leads to the development of a specialized organ where nitrogen fixation takes place – the root nodule. Nodule organogenesis is a process that occurs only under nitrogen-limiting conditions and that is promptly terminated when an inorganic source of nitrogen is sensed. Hence, components of nodulation pathways must surely overlap that of the nitrogen sensing/signaling pathways. Although some of the key players within nitrogen signaling pathways have been revealed in recent years, we are still far from completely understanding this complex network. I am currently interested in investigating how nitrogen signals are integrated with plant developmental processes, at the molecular level, particularly during the Legume-Rhizobium interaction. Within this context I aim to understand the role and regulation of prokaryotic GSI-like proteins in M. truncatula..
5. Plant-Obligate biotrophs interactions
Objectivos»Goals: A key question that has been poorly addressed is how plant obligate parasites adapt to new hosts? Recent findings lead us to hypothesize that heat shock protein 90 (HSP90) may be involved in such process. For instance, during stress HSP90 appears to link pre-acquired genetic diversity with fast appearance of new traits in eukaryotes. In addition, HSP90 appears to function as a virulence factor in human protozoa parasites. The goal of this research was to investigate the impact of cytoplasmic, and especially apoplastic HSP90s, on adaptation of obligate parasites to new plant hosts, to be specific, to investigate whether the proteins promote virulence and/or function as a buffer for genetic variation. To explore this topic I used the model plant Arabidopsis thaliana and the obligate parasitic oomycete Albugo laibachii. To reach my objective, I addressed the following questions: 1) How are HSP90s secreted? 2) What is the relative contribution of cytoplasmic versus apoplastic HSP90 to virulence? 3) What is the impact of stress, and hence the impact of HSPs to host shifts? .

Outras informações relevantes
My research expertise relates mainly to nitrogen metabolism in legumes with emphasis on nitrogen assimilation in root nodules and developing seeds.
Most of my work has focus on clarifying aspects related to the role and regulation of glutamine synthetase isoenzymes in Medicago truncatula.
To accomplish my goals I have combined different disciplines such as physiology, molecular biology, biochemistry and structural biology and have successfully published my work in scientific journals of high impact.
During my research career I have also engaged on different projects related to host-microbe interactions, which have broadened my research skills in ecology, microbiology and immunology.
As a post-doc researcher I used the Arabidopsis thaliana-Albugo laibachii pathosystem to understand the life style of plant biotroph pathogens and as an undergraduate intern I studied the B-cell response of mice models to the parasite Neospora caninum.
Other interests include the development of sustainable and effective methods of crop production and actions of science communication for general public, which included participation as a teacher in children educational activities promoted by Porto Municipality.
