Área de actividade científica - Glutamatergic Synapses - Formation and regulation of synapses Neurons have a complex morphology,
with branched dendrites exhibiting thousands of synapses, a specialized region where communication between neurons occurs.
We are interested in understanding how these connections between nerve cells are formed and how they are modified with experience.
The ability of synapses to change their strength is thought to be the cellular correlate of learning and memory, and synaptic
dysfunction is correlated with several neurodegenerative diseases. We focus on excitatory glutamatergic synapses, and
study their regulation and formation from a cellular and molecular biology viewpoint. We use a combination of techniques
like primary neuronal cultures, biochemistry, molecular and cellular biology to address these questions. Technical
skills and competences Cell Biology Hippocampal cells primary cultures; HEK-293 cell line; Transformation of bacteria cells;
Plasmid transfections in primary cultures using the calcium phosphate method; Plasmid transfections in cell lines using both
the calcium phosphate method and lipofectamine; SDS-polyacrilamide gel electrophoresis; Western blotting; Protein concentration
determination using the BCA method; Polyacrilamyde gel staining with Coomassie blue; PVDF membrane staining with Coomassie
blue; Pull-down and immunoprecipitation assays; Immunocytochemistry; Fluorescence microscopy Molecular Biology PCR and
RT-PCR; Plasmid cloning, in situ mutagenesis and deletions, RNA extraction and purification and DNA purification using kits
for mini, midi and maxi prep and homemade solutions; Recombinant GST fusion protein and His-tagged protein expression and
purification from E. Coli; DNA analysis and purification from agarose gel using kits; Preparation of E. coli competent
cells Computer skills and competences: Experience in laboratory programs such as GraphPad Prism (statistical analysis),
Endnote (References database), ImageQuant (western blot quantification), Vector NTI (Molecular Biology program), AxioVision
(Microscope software) and ImageJ. Experience also with Microsoft Word, Power Point and Excel. Complementary training
