The Handbook of Stress and the Brain focuses on the impact of stressful events on the functioning of the central nervous system;
how stress affects molecular and cellular processes in the brain, and in turn, how these brain processes determine our perception
of and reactivity to, stressful challenges - acutely and in the long-run. Written for a broad scientific audience, the Handbook
comprehensively reviews key principles and facts to provide a clear overview of the interdisciplinary field of stress. The
work aims to bring together the disciplines of neurobiology, physiology, immunology, psychology and psychiatry, to provide
a reference source for both the non-clinical and clinical expert, as well as serving as an introductory text for novices in
this field of scientific inquiry.
Part 1 addresses basic aspects of the neurobiology of the stress response including the involvement of neuropeptide, neuroendocrine

and emotionality.
Part 2 treats the complexity of short-term and long-term regulation of stress responsivity, the role of stress in psychiatric

"Stress shifts the response of the bed nucleus of the stria terminalis to an anxiogenic mode", European Journal of Neuroscience 36, 10: 3396 - 3406. The bed nucleus of the stria terminalis (BNST) is critically implicated in anxiety behavior and control of the hypothalamus
pituitary adrenal axis. Having previously shown that chronic stress triggers dendritic/synaptic remodeling in specific nuclei
of the BNST, we characterised the pattern of activation of neurons within different regions of the BNST under basal conditions
and after an anxiogenic stimulus in control and stressed rats. Under basal conditions, stressed, but not control, animals
displayed increased cFOS expression in the dorsomedial nucleus and decreased activation of the principal nucleus. This pattern
resembled that observed in controls that had been exposed to the anxiogenic stimulus. Subsequent analysis of various BNST
subnuclei revealed differential patterns of gene expression in controls and stressed animals. We found decreased levels of
corticotropin-releasing hormone 1 receptor mRNA expression in the dorsomedial and fusiform nuclei, and a global increase in
the levels of corticotropin-releasing hormone 2 receptor in the principal nucleus. In addition, we found subnuclei-specific
increases in GABAA and NR2B receptors in stressed animals, which suggest changes in the GABAergic and glutamergic innervation
of the BNST. Importantly, these findings were associated with increased anxiety-like behavior and impaired control of the
hypothalamus pituitary adrenal axis in stressed animals. In summary, these data reveal that chronic stress shifts the pattern
of response of the BNST to an anxiogenic mode and provide new information on the underlying mechanisms of the stress-induced
hypercorticalism and hyperanxious status.

"Programming Effects of Antenatal Corticosteroids Exposure in Male Sexual Behavior.", The Journal of Sexual Medicine, 10000: 10000 - 10001. Introduction. Brain regions implicated in sexual behavior begin to differentiate in the last trimester of gestation. Antenatal
therapy with corticosteroids is often used in clinical practice during this period to accelerate lung maturation in preterm-risk
pregnancies. Clinical and animal studies highlighted major behavioral impairments induced later in life by these treatments,
especially when synthetic corticosteroids are used. Aim. To evaluate the implications of acute prenatal treatment with natural
vs. synthetic corticosteroids on adult male rat sexual behavior and its neurochemical correlates. Methods. Twelve pregnant
Wistar rats were injected with dexamethasone (DEX-1 mg/kg), corticosterone (CORT-25 mg/kg), or saline on late gestation (pregnancy
days 18 and 19). Following this brief exposure to corticosteroids, we assessed the sexual behavior of the adult male progeny
and subsequently associated these behaviors with the levels of catecholamines and mRNA of dopamine and androgen receptors
(AR) in brain regions relevant for sexual behavior. Main Outcome Measures. Sexual behavior of adult male offspring was assessed
by exposure to receptive females. This was associated with serum testosterone levels and levels of catecholamines (determined
by high-performance liquid chromatography) and dopamine and AR mRNA expression (real-time polymerase chain reaction [PCR])
in brain regions implicated in sexual behavior. Results. Prenatal DEX exposure resulted in a decreased number and increased
mounts and intromissions latencies in adulthood. These findings were associated with decreased levels of serum testosterone
and increased hypothalamic expression of AR mRNA. DEX animals also displayed lower dopamine levels and higher dopamine receptor
mRNA expression both in hypothalamus and nucleus accumbens (NAcc). The milder phenotype of CORT animals was associated only
with decreased dopamine levels in NAcc. Conclusion. Antenatal corticotherapy programs adult male sexual beha.

"Invivo assessment of gastrotomy closure with over-the-scope clips in an experimental model for varicocelectomy (with video).", GastrointestEndosc 70, 6: 1137 - 1145. Abstract BACKGROUND: Gastrotomy closure remains the major limiting factor for human translation of transgastric surgery; the
over-the-scope clip (OTSC) system was proposed as a possibility for this purpose. Transgastric access is good for a pelvic
approach, making varicocelectomy a possible indication for natural orifice transluminal endoscopic surgery (NOTES). OBJECTIVE:
To evaluate the reliability of the OTSC system in vivo after transgastric testicular vessel ligation (varicocelectomy model).
DESIGN: There were 3 experimental groups (5 animals in each): groups 1 and 3, gastrotomy dilation up to 18 mm, surgery was
performed with a double-channel endoscope; group 2, gastrotomy dilation up to 13 mm, surgery was performed with a single-channel
endoscope. SETTING: Surgical Sciences Research Domain, Life and Health Sciences Research Institute (ICVS), School of Health
Sciences, University of Minho, Braga, Portugal. INTERVENTIONS: Bilateral testicular vessel ligation by transgastric access.
Gastrotomy closed with the largest version of OTSC system (12 mm): a single clip in groups 1 and 2, and 2 clips in group 3.
Animals were monitored for 2 weeks, killed, and submitted for necropsy. MAIN OUTCOME MEASUREMENTS: Adequacy of closure and
healing after the use of the OTSC system. Statistical analysis. RESULTS: Vessel ligation was easily achieved in all groups.
Although differences in the complication rate did not reach statistical significance (P = .099), there was a clear tendency
for a better prognosis in groups 2 and 3 than group 1. In fact, only 2 animals from group 1 had complications related to incomplete
gastrotomy closure. LIMITATIONS: Small number of animals per group; nonrandomized study. CONCLUSIONS: The OTSC system was
shown to be easy and efficient for gastrotomy closure in a survival experimental model of varicocelectomy, when correctly
matching the gastrotomy size with the clip size and/or number.

"Antinociception induced by chronic glucocorticoid treatment is correlated to local modulation of spinal neurotransmitter content.", Molecular Pain, 5: 41 - 52. While acute effects of stress on pain are well described, those produced by chronic stress are still a matter of dispute.
Previously we demonstrated that chronic unpredictable stress results in antinociception in the tail-flick test, an effect
that is mediated by increased levels of corticosteroids. In the present study, we evaluated nociception in rats after chronic
treatment with corticosterone (CORT) and dexamethasone (DEX) in order to discriminate the role of each type of corticosteroid
receptors in antinociception. Results Both experimental groups exhibited a pronounced antinociceptive effect after three
weeks of treatment when compared to controls (CONT); however, at four weeks the pain threshold in CORT-treated animals returned
to basal levels whereas in DEX-treated rats antinociception was maintained. In order to assess if these differences are associated
with altered expression of neuropeptides involved in nociceptive transmission we evaluated the density of substance P (SP),
calcitonin gene-related peptide (CGRP), somatostatin (SS) and B2-¿-aminobutiric acid receptors (GABAB2) expression in the
spinal dorsal horn using light density measurements and stereological techniques. After three weeks of treatment the expression
of CGRP in the superficial dorsal horn was significantly decreased in both CORT and DEX groups, while GABAB2 was significantly
increased; the levels of SP for both experimental groups remained unchanged at this point. At 4 weeks, CGRP and SP are reduced
in DEX-treated animals and GABAB2 unchanged, but all changes were restored to CONT levels in CORT-treated animals. The expression
of SS remained unaltered throughout the experimental period. Conclusion These data indicate that corticosteroids modulate
nociception since chronic corticosteroid treatment alters the expression of neuropeptides involved in nociceptive transmission
at the spinal cord level. As previously observed in some supraspinal areas, the exclusive GR ac

"A Behavioral Characterization on the Motor and Emotional Deficits of the 6-OHDA Parkinson Disease Rat Model", Trabalho apresentado em 12th Meeting of the Portuguese Society for Neurosciences, In 12th Meeting of the Portuguese Society for Neurosciences, Lisboa. Parkinson’s disease (PD) is a neurodegenerative condition affecting the dopaminergic system, causing the emergence of both
motor and non-motor complications. The development of animal models mimicking this condition brought new insights regarding
the neuropathologic mechanisms of this disease, and endowed us with suitable tools to develop new therapeutical strategies.
One of the most widely used models is the unilaterally 6-OHDA-lesioned rat model, characterized by a biased rotatory behavior
after apomorphine/methamphetamine challenge. However, little is known about other behavioral features of this model, particularly
at the emotional/cognitive level. Importantly, this model has been extensively used to assess the therapeutical relevance
of novel treatments, particularly at the motor level. The improvement of the behavioral characterization of this model could
reveal additional features besides the motor deficits, which could in the future be useful to address treatment-related improvements
in a multidimensional fashion.
Therefore, the objective of the present work was to pursue a more thorough behavioral characterization of this model, particularly
at the emotional and cognitive levels. We believe that our findings can be highly relevant for the validity of this model
in PD regenerative medicine.
Wistar-Han rats were unilaterally injected with 6-OHDA or saline in the medial forebrain bundle (coordinates related to bregma,
AP= -4,4mm; ML= -1,0mm; DV= -7,8mm). Behavioral assessment was done using: rotorod, elevated plus maze (EPM), acoustic startle
(AS), Morris water maze (MWM), open field (OF), forced swimming test (FST), sucrose preference test (SPT) and rotameter (apomorphine-derived
rotatory behavior assessment). Tyrosine hydroxylase immunohistochemistry was performed to assess the effectiveness of the
lesion.
Lesioned animals displaying apomorphine induced rotatory behavior had severe dopaminergic degeneration. In this group of animals,

e permitir uma transição menos abrupta foi idealizado um curso de iniciação à Anestesiologia Clínica – InAnestesia. O curso
foi dirigido aos internos do primeiro ano de especialidade, durante a primeira semana de trabalho e pretendeu criar um ambiente
de introdução à prática da Anestesiologia. Os aspectos formativos do curso, englobaram fundamentos teóricos, básicos à prática
da Anestesiologia, apresentados sob a forma de sessões teóricas, e fundamentos práticos com recurso a simuladores, onde os
internos puderam aprender e treinar gestos clínicos da prática diária dos anestesiologistas que, na globalidade, consideramos
essenciais para os primeiros passos na especialidade.

also shown cognitive and molecular changes induced by GA exposure. The aim of this project is to determine the effects of

dopaminergic after GA exposure. For the purpose we will expose rodents to GA to try to assess changes in cognition processes
related to cortico-basal ganglia network. |

underlying anxiety, 2010. Dissertação (Neurociências) - Universidade do Minho, Bolseiro(a) de Fundação para a Ciência e a Tecnologia (Orientador). Anxiety disorders affect a large portion of the world population and are frequently long-lasting and debilitating. Chronic
stress plays an important role in their etiology. Several studies have shown that animals exposed to chronic unpredictable
stress (CUS) present anxiety-like behavior correlated with hypothalamic–pituitary–adrenal (HPA) axis dysfunction and structural
changes in the brain, principally in limbic structures.
The mechanisms by which chronic stress alters central circuits that mediate anxiety-like emotional behavior are still largely
unknown. Substantial evidence has implicated the bed nucleus of the stria terminalis (BST) in the regulation of the HPA axis,
acting as a relay nucleus between limbic structures and the hypothalamic paraventricular nucleus (PVN). This is enhanced by
previous results showing a hypertrophy of dendrites and spine sprouting in the BST anterodivision after CUS exposure. Nevertheless
it is not known if the stress-induced structural and behavioral changes are associated with an altered pattern of activation
in the BST. To address this question, electrophysiological studies will be conducted in stressed and control animals. A CUS
paradigm will be used to induce anxiety-like behavior in rats, which will be evaluated in three different tests: open field,
elevated plus maze and acoustic startle. Electrophysiological proprieties of single cell units in dorsomedial/fusiform and
principal BST subnuclei of both stressed and control animals will be recorded in basal conditions. Afterwards BST spontaneous
and sonorous-evoked activity will be recorded immediately after manipulation of central nucleus amygdala, in order to see
the role of amygdala inputs over BST.
